Phase Contrast Microscopy
Phase Contrast Microscopy is employed to determine airborne fiber concentrations using NIOSH Method 7400. This methodology involves drawing a calculated quantity of air through a mixed-cellulose ester filter, which is housed in a 25mm open-faced cassette. A wedge of the filter is mounted onto a microscope slide using an acetone-triacetin clearing process and a manual count of fibers, which are greater than five microns in length and possess a length to width aspect ratio of at least three to one, is performed. A simple calculation determines a fiber per cubic centimeter of air concentration.
